Penryn ward encompasses the historic town of Penryn itself, located at the head of the Penryn River, a tributary of the Carrick Roads. The area includes the town's dense network of steep, narrow streets and granite buildings that reflect its medieval origins as a stannary town. The ward extends to incorporate surrounding residential areas and stretches of the river's edge, including the working wharves at Town Quay.

The University of Exeter and Falmouth University maintain a significant shared campus here, known as the Penryn Campus, which is a major presence within the ward.
